I think that the point of this movie is to parallel the Nazis to the Philistines, who were the biblical enemies of the Hebrews.

I am not sure if the ark could be opened by anyone, ritual or not. In a passage from Leviticus, God tells Moses to warn his brother Aaron about not going in to the ark at all times. So we know that not even he was safe. The Bible also says that David was afraid of carrying the ark home.
