I thought the funniest part was when Schmidt wrote his first letter to Ndugu and got really worked up about being replaced at his job. It was the hardest I've laughed during a movie in a while.

The singing at the wedding - LOL The whole ceremony, the speeches, the "sensitive" vows: "I shall love you every day..I mean all 24 hours.. all 86,400 seconds" The prime rib carvery, the drunken Best Man speech. =)
A few have mentioned Warrens first letter to Ndugu and him getting angry about his job. I do like that bit, however, the part that made me laugh was also whilst writing the first letter, he is giving some general information and then he randomly says: "I have an older brother who lives with his wife, but sadly he has just lost a leg to diabetes"
That made me laugh when he said that, you can just imagine the Nun reading the letter to Ndugu and thinking "Oh! thanks for sharing that!"
The part in the letter about the brother losing his leg is funny. After that my bf said, now that's way too much information.
For my money, though, and I'm not even sure why it's so funny, is the scene in which we first see Randall, coming out of the waterbed store. His mullet, the thin hair that he's plastered to his head, those horrible mismatched clothes. Even the cars in front of the store are hysterical--a red Pinto station wagon and an old El Camino (which I think is Randall's).
Really, a great scene, and one of the best opening scenes I've ever seen. In only half a minute you can clearly see Warren's character better than some characters in other movies can be seen at the end of the movie.
I need to go back and watch this again. I didn't find anything funny at all within this movie or about this movie. It was being sold in the comedy section, so I bought it on a whim expecting a light hearted comedy. This movie was the complete opposite, which probably led to me being so let down by it. Maybe if I just approach it more as a Drama than a comedy, I might enjoy a little more.
If you haven't seen 'Sideways', try looking at that first. It is also somewhat of a dark comedy but I think the humor is a little more easy to spot. Perhaps it'll help get a sense of where Alexander Payne comes from with his ironic view of life and the people that struggle to get through it.
Just a suggestion.

no one came close to mentioning this part, but the funniest part by far to me was when he was recouping in bed, reading "encyclopedia brown gets his man" and then flipping to the back to reveal the solution to the childish mystery and how he grins and shakes his head, as if he never saw it coming. genius.
The peeing performance reminded me of "Wolf" where he moves about in the same way and ends up peeing on James Spader's shoes ("just marking my territory").
Funniest scene has to be Warren looking at Randall's awards for "participation" and certificate for completing a 2 week course in electronics, hehe!
*SPOILER ALERT* Another subtly funny scene occurs after Warren barrels out of the trailer park after he comes on to the wife of the guy who went out for more beer. The guy sees Warren leaving in his RV,and waves goodbye with a total WTF? face.
A funny part not included in the movie, but is on the DVD deleted scenes, happens right after the trailer park incident. It is the next morning and the cops arrive with Schmidt's van parked illegally on the side of the road. The cop wakes Schmidt and tells him to move. Schmidt explains he was tired and had to pull over to sleep. The copper says 'there was a trailer park 12 mile back, how could you not have seen it?'
Well, it's a pity that you didn't have the opportunity to have Dubchek, Ceuasescu, Pol Pot, Honnecker or Jaruzelsky for a president. It might have been a revealing experience, a lot of understanding would have been opened for you...
