The end of DEFENDING YOUR LIFE (which is otherwise a quite lighthearted film) always gets me teary! It's a combination of the music and all that's happening.
The lead character is in Purgatory, realizing he didn't take enough risks in his earthy life, and may be sent back for reincarnation.

I enjoyed that movie and the final scene, but I never really sensed any chemistry between Albert Brooks and Meryl Streep.

Well, the whole movie is very slight, IMO. It's true you don't feeel an electric connection or anything between the two...but it sort of works (at least, well enough) because who wouldn't love Meryl Streep? And she seems so kind in it, you can buy she'd love him (along with everyone else).
I do see what you're saying, tho. They're not exactly star-crossed lovers.

Last goodbye moments, where you know the characters are probably never going to see each other again, but they have to part. That stuff gets me every time.

Turn it around the opposite way..... Reunions after many years bring me to tears as well. I love reunions. I get very emotional.

I'd have to say reunions, done emotionally, are very heart-wrenching. Sadly, most reunion movies/moments go for the comedy more than anything.
