a central character is a loner/misfit desperately searching for love/companionship but ends up losing it when rejected. The ending should be bleak and depressing.
Any suggestions?

really similar all of them, espcially since they are all horror films with black comedy infused, May is still the best of the bunch.
I would have to agree with some of the other posters and also recommend One Hour Photo, Taxi Driver and Roman. Roman was directed by Angela Bettis (who played May) and she said herself it is a similar movie to May, but from the point of view of a male.
Though I know many people like it, I wouldn't recommend Spiral. I personally found it boring for the most part and didn't like Joel David Moore's acting.
You remind me of a white crayon... totally useless.
My pick was Excision but several people picked that movie. So I will go with this one.
Bug (2006)
It may not exactly be the type of movie requested, but for me, a perfect companion piece to May, was American Mary http://www.imdb.com/title/tt1959332/?ref_=fn_al_tt_1. In both movies, the protagonist goes through an incredible growth through their struggle for survival. By the end of each, they are both nearly different people.
