What about films from the 60s that deal with racial themes like In The Heat of the Night and Guess Who's Coming to Dinner? Aren't they "woke"?

That's fair. I've never seen either, but I'm vaguely familiar with them. This would make an interesting topic for research. What really might be considered the First Woke Movie?

No Way Out and Storm Warning, both from 1950, also deal with racism and could be considered woke.
