The first time I recall hearing this was in The Hangover. Recently it is being used so much. I don't find the line funny as it doesn't make any sense. Am I alone in this opinion?

Can you cite five instances within this year of characters saying the same phrase? It should be easy if it's recently being used so much.

It's slang and obviously not literal. It's a short, simple way of telling someone you don't care what they have to say and to go fuck themselves.

Same. In addition to insulting the guy a woman might say this to, it often confuses the dude and throws him off momentarily as well. Double whammy!
