I don't think Peacemaker's "that's racist" comment was meant to get a ton of laughs. The big laugh was supposed to be just seeing him in tighty-whities.
The line is really just a characterization of Peacemaker as a uninformed idiot; Bloodsport was obviously not being racist. On top of Peacemaker's stupidity, he was shown to be easily manipulated (his instant and total loyalty to Waller) and incredibly self-righteous (not caring how many men, women, or children he needed to kill for peace). The "that's racist" line is part of the set up to Peacemaker's confrontation with Flag.
