I dont know about biggest laugh but when Will confronts the douchey hippy traveller guy, the whole cinema cheered and clapped (a reaction very unusual for an British cinema!)
shareThe poo scene and the dog licking Neils balls. It was a great crowd, I have not heard people laugh that much and loudly for a long time. This man laughed at a scene no one else did that made everyone laugh.
shareEither the *beep* to the face or Will singing "The First Time Ever I Saw Your Face" got the biggest laugh from me, my mates and the cinema. Once again as is the standard, the sequel is not as good as the first one but I still pissed myself through most of this one! Can't wait til it comes out on DVD

Laughed hard at the scenes with Uncle Bryan, as an Aussie that was no exaggeration, some blokes are that frightfully 'ocker' very well played by Dave Field :)
