Based on a quick Google search I can't find anything that says Da Vinci was ever in Spain and during the time frame of the movie I think Da Vinci was in Rome. Ezio might appear there was a portable game called Assassin's Creed: Discovery where Ezio went to Spain during the same time period as the game so he could have a cameo or something.
