The idea of "nuclear mutants" was one a very common science fiction trope. These days we mainly see it in superhero/villain origin stories, but it was once popular with top writers like Philip K Dick and Harlan Ellison. Back in the day, radiation was assumed to mutate existing life into whole new species or monsters overnight, or at most a generation.

I assumed it was not the radiation but the confinement in a dark cell that made the escaped patients go mad and attack the others.

If a person is "burned" that badly by radiation, they would die from radiation poisoning, unless getting serious medical attention.

Well, when you get bad radiation poisoning it can scramble your brain up a bit, but that's usually a lethal dose so you'd live like a week or so
