I saw it several times in several different theaters. Each time was a little different in the number of audience members and the various scenes that got small and big laughs. is there something in particular you were curious about? I could list many things that I remember about the audiences that I was a part of. If you could narrow it down for me i'll be happy to share my experiences.
My theater was very quiet for what I can remember. I saw it in a theater in SoHo, Manhattan.

I only saw it once. People nervously laughed at 3 (4?) uncomfortable moments:
-Sam telling the story of her first sexual encounter
-The Rocky Horror Picture Show
-When Patrick kissed Charlie
But, apart from those scenes, good audience...
I saw it on a whim, a Monday, not 20 ppl in the place. It was one of the more memorable theatrical experiences actually because I think its emotional depth & breadth caught everyone off guard- 180 from its cliched high school dramedy marketing.
While no one seemed quite sure where it was going, & while the laughs were genuine all over the audience, there were of course its countless instances where siht gets real, probably for different viewers at different instances. For me the first biggie was revealing his friend's suicide, & the next was the 'Something' LP reveal. The first told me who Charlie was & the second told me his perception of his friends, notably Sam, & how sincere & mature those relationships were becoming.
I went back that Thursday. I'd never in my life returned to watch a movie so soon, I craved the experience & also felt like giving it some more business as if one more ticket was like being able to vote twice.
That viewing was fascinating unto itself, seeing new ppls reactions to its lines & emotional reveals. There was a female duo a few aisles down who were laughing at the characters a lot rather than with, & completely blown away by Charlie's spin the bottle inner dialogue ('...kept fantasizing once of us dying of cancer') as if they were about to quit on the movie. It was interesting how abruptly they sat up with its pay off.
