Before Frank and Jon go on stage at sxsw Frank decides to change into a dress and comes out of changing room saying "Let's *beep*".
I never understood the point of this (if there even was any). Any insights?

Well he was stressing out pretty hard at that time. I think the dress and makeup was another "layer" for him to hide behind.

According to the writers' commentary on the DVD, this is Frank trying to be Britney Spears - his idea of what a pop star is "like".

I took it as Frank trying to be the Clara to Jon's Frank. Or perhaps trying to imitate the new girl/guy duo fad in music these days.
