(Following the events of the last film, Priest, Priestess, Hicks, Lucy, and Lucy's drifter friend, Kate, are now on the run, hunted by both vampires and humans. Monsignor Orelas still has faith in Priest, and tries to negotiate with Monsignor Chamberlain to spare their lives. However, Priest's mentor, veteran priest, Father Garcia conspires with Chamberlain to find the Salesman, who has picked up where Black Hat left off by creating an entire legion of human vampires. After locating the dreaded creature, Garcia's lust for immortality forces him to promise the Salesman the city and the holy church if he gives him a taste of what he has become. When Monsignor Orelas discovers their plot, he is betrayed to Salesman's army and is drained of his blood. Father Garcia and Monsignor Chamberlain are then turned into vampires and begin turning several priests and civilians. Priest has been able to fight off all human vampires, creature vampires, and bounty hunter priests he used to call brothers. However, his pupil; a young priest once named Jonah Vale, has been turned into a vampire and sent to kill them. The Salesman and his right hand, Benoit, were reluctant to share their power with such a greedy partner, and soon conspire themselves to destroy Father Garcia and Monsignor Chamberlain. Amazingly, two mercenaries turned vampire slayers, Danson and Carmine, escape the city to find Priest and his gang to let them know what happened. Priest, trying to evade his protege, returns back to his beloved city in order to stop The Salesman and those who have betrayed their God and turned to the Devil for salvation.)

Especially the way that the film ended with Priest throwing the beheaded vampire head into the council chambers suggesting that the war is far from over.
