It is pretty funny, but I don't understand why this line seems dubbed in. Maybe after they wrapped shooting they though "OH YOU KNOW WHAT SHE SHOULD HAVE SAID"

shareIt was Will, right after Gwen reveals to him that she's not Royal Pain's daughter, she is Royal Pain. Regarding the OP, MetMacFan is right. It's only funny because it's a clever reference. I wouldn't have understood it if I didn't have the assumed knowledge. In the context of the movie itself, I'd say the funniest to me were "I went through puberty twice for this?!", "Surprised? So am I!" and the whole dialogue between the sidekicks and Warren when they sit down at his table. But my favourite line is undoubtedly the last line. I agree with steven-digby, it's one of the most apt and original ending lines of any movie.

In my opinion, the best line in the movie is:
When Royal Pain is explaining her backstory, Stitches calls her "Daddy's little girl", and she yells "told you never to CALL ME THAT!"
To me, that line never gets old.
