Do I need to see Blade to understand this?
I'm more interested in seeing this because of Guillermo del Toro over anything else. Would I understand it if I didn't see the first one? Is the first one just worth seeing no matter what?

shareIt's not nessecary to see the first one, unless you want to know the fate of Blade's mother. The main villain of the first one is briefly referenced. Part 2 picks up where the first one left off, Blade mentions that in this movie, he also recaps his life story leading up to the events of the second film.
Reboot, restart or re-imagining is another word for remake

Not really. There's only one reference that's not spoon fed to you in the opening and throughout the film
