In the film, there was a joke in supermarket scene which we don't know the answer to:

"What's a hooker say after 12 hours on the job?"

Does anyone know the punch-line for the joke? Any help would be appreciated!

The joke (at least, in the movie) is basically:

"What's a hooker say after 12 hours on the job?"
(spits milk out)

The milk is supposed to represent the semen that's gotten into the hooker's mouth from 12 hours of blowjobs.
