Don't take this the wrong way but I don't want Stryker to be in the new movies since in the comics he actually had little to do with Wolverine's origin. I want Professor Oyama to be the main guy responsible for Wolverine's skeleton. And for his daughter Yudigo to become the antagonist Lady Deathstrike.
