This whole story feels like a Tragedy with a bunch o people who are, essentially, wasting their lives accomplishing nothing. I smiled a few times at the jokes, but overall the movie left me feeling depressed. (Much like another tragedy, Death of a Salesman.)
It's not really a comedy OR a tragedy. There are some crazy things that the protagonists experience that can seem funny, and things that happen to people BECAUSE of the protagonists that can seem tragic, but this story escapes definition.
It's supposed to just be 'gonzo' reporting, which means getting as deep into the story as you possibly can, regardless of the consequences.
An important thing to recall is that Hunter S. Thompson admitted that he had no idea how much of the story actually happened, and how much was made up/hallucinated.

I couldn't watch this movie because of all the vomit scenes. Really gross and stupid.

It sums up the feeling of Hunter's generation as the the sixities ended and the seventies began. The nihilism of a generation under Nixon and the fantasy escape that Las Vegas personified (anything goes). It is a bit depressing, but ultimately consider that Duke is Hunter, he is a writer and he is conveying the complexities and depths of the madness, not just mindlessly indulging - although that dilemma is one central to the film and the characters talk about it.
It is a stinging critique of America and of both the totalitarian fundamentalist regime of Nixon and the conservatives propping it up as well as the hippies and their hedonistic nihilism. And amazingly it strikes a chord now and probably always will.

I don't think the story is really telling you, one way or the other, what to feel about the behavior and lifestyle of the characters.
They do what they do, and you can think what you want of them.
