Quite a stupid line. Darby gives Gray instructions that will ultimately lead him to her hotel room, and when he shows up at her room, she asks: "Did you follow my instructions"? How do you think he got there?
If she was afraid that he was someone else than Gray, she would have asked "Who are you", but since she asks him about the instructions it`s obvious she knows this is Gray.

I took it more to mean that she didn't want him to just come straight there from point A to point B because he could be more easily followed. Apparently she told him a bunch of things to do that would hopefully lose anyone following him. If he chose to ignore those steps and just come directly that would make her more worried and scared. I think she just was asking to ease her mind and confirm that he took all of these extra steps that would lose those that could be following him.
