Why does the passage have to be spoken in German? What difference would it make if it was spoken in English or any other language.

That's the language the diary containing the passage was written in. That's why the kids had to find a person who spoke German. I don't think it had to be read aloud in German, but it was written in German, so, eh.
Interesting tidbit: in Bram Stoker's novel, Van Helsing was actually Dutch, and they don't speak German. They speak, well, Dutch, so Van Helsing's diary should've actually been written in Dutch.
