I think it was a bigoted (note: this is a grindhouse film, so things aren't politically correct) statement at the time when Japan opened their land to foreigners, post-World War II. Not many Japanese liked it when their women were with African-American men, after being exposed to racist views from whites. Fortunately, the younger generation's more acceptable there.
As for the scene, it's pretty cruel but funny.
