I just saw this again recently. It occurred to me, the ending id kind of like King Kong don't you think (or maybe I should say here it would be Queen Kong)? He climbed the tower holding injured Frank-N-Furter

There are many deliberate references to other movies and the ending, does reference King Kong. Other references, aside from the Frankenstein theme, include the Wizard of Oz, Dr Strangelove and Night of the Hunter.

Frank sings "Whatever happen to Fay Wray?" Fay Wray is the actress in the original 1933 version of King Kong.
So yes it is a nod to King Kong.
RHPS refers to many many movies. There's hardly an original idea it in it just blends all of them in an unique and wonderful way.
