With all the rehashing/rebooting of old movies, I think Fantastic Voyage would be an interesting remake. If anything, just updating the technology used in the story can be intriguing.

You should check out Innerspace (1986)
Its roughly the same idea.

Someone posted an article last year that David Goyer and James Cameron had been working on it.
