It's just a joke. It doesn't really mean much. The premise of the relationship between the fictionalized John and the fictional manager is that they're constantly in a sort of war with each other: the manager being the "adult" who tries to keep order, and Lennon being the class clown/cut-up who never takes orders. It's just part of that context - it doesn't mean anything else.
The world of the movie differs from the real world, of course. John and Paul each have mothers, and John does not appear to have a wife, just to mention a few.
The creepy line "John, put those girls down or I'll tell your mother on you" always sounds wrong.
